Identify the correct sequence which are involved in DNA fingerprinting.
A. Separation of DNA by electrophoresis
B. Isolation of DNA
C. Transfer of separated DNA to synthetic membrane
D. Hybridisation using labelled VNTR probe
Choose the correct answer from the options given below:
Answer & explanation
Correct answer: option 2
The correct answer is Option (2) → B, A, C, D
DNA fingerprinting compares the repeat sequences of DNA (VNTRs) of different persons. The method used earlier was Southern blot hybridisation.
B. Isolation of DNA — first. The DNA is taken out of the cells. It is then cut with restriction endonucleases.
A. Separation of DNA by electrophoresis — second. The cut fragments move through a gel and separate by size.
C. Transfer of separated DNA to synthetic membrane — third. The separated fragments are blotted on to a nitrocellulose or nylon membrane. Only then can a probe be applied.
D. Hybridisation using labelled VNTR probe — last. The labelled probe sticks to the matching fragments. They are then seen by autoradiography as bands.
